Holmes County is a county located in the Mississippi Delta region of the U.S. state of Mississippi. As of 2000, the population was 21,609. It is named in honor of David Holmes, the first governor of the state of Mississippi. Its county seat is Lexington.

The racial makeup of the county was 20.47% White, 78.66% Black or African American, 0.12% Native American, 0.15% Asian, 0.07% from other races, and 0.52% from two or more races. 0.90% of the populations were Hispanic or Latino of any race.
